The Pattern of Genetic Diversity of Palm Oil (Elaeis Guinensis Jacq) DXP 540 NG Varieties from PPKS Using RAPD Markers (Random Amplified Polymorphic DNA)

Introduction

The genetic diversity of palm oil (Elaeis Guinensis Jacq) is a crucial aspect of understanding the species' adaptability to environmental changes and resistance to diseases. The DXP 540 NG variety, originating from the Palm Oil Research Center (PPKS), has been the subject of various studies aimed at analyzing its genetic diversity. This study aims to investigate the pattern of genetic diversity in the DXP 540 NG variety using RAPD (Random Amplified Polymorphism DNA) markers. The study was conducted at the Agricultural Biotechnology Laboratory, Faculty of Agriculture, University of North Sumatra, Medan, between March 2018 and January 2019.

Materials and Methods

A total of 20 samples of palm oil leaves from the DXP 540 NG variety were successfully isolated for genetic analysis. The samples were analyzed using RAPD markers, which are a type of PCR (Polymerase Chain Reaction) technique that amplifies specific DNA sequences. The RAPD markers used in this study were designed to detect polymorphic bands, which are indicative of genetic variation. The size of the amplified DNA fragments ranged from 446 to 1574 bp. The genetic distance and dendrogram formation were calculated using Darwin 6.0 software.

Results

The results of the analysis showed that 19 out of 20 samples were successfully processed using genetic analysis software, while 1 sample did not meet the specified standards. The total number of polymorphic bands produced was 14, with all bands being polymorphic (100%). The size of the amplified DNA fragments ranged from 446 to 1574 bp. The calculation of genetic distance and dendrogram formation revealed that the 19 individuals analyzed were divided into 3 main groups. The molecular diversity of the DXP 540 NG variety was found to be 49.79%, with a dissimilarity coefficient ranging from 0 to 0.75.
