Introduction

Red beans (Phaseolus vulgaris L.) are a rich source of vegetable protein, containing a protein content of 22.1%. Research Methods

The isolation process begins with red bean extraction using sodium hydroxide (NaOH) until it reaches pH 8. Furthermore, protein deposition is carried out with the addition of 2N HCl to pH 4 and CH3COOH 2N to pH 4.5. Protein content is measured using both methods, namely Kjeldahl and Bradford, to get accurate results regarding the protein content in isolates.

Materials and Methods

Red beans (Phaseolus vulgaris L.): The red beans used in this study were obtained from a local market.
Sodium hydroxide (NaOH): Used for alkaline extraction of red beans.
Hydrochloric acid (HCl): Used for protein deposition with a concentration of 2N.
Acetic acid (CH3COOH): Used for protein deposition with a concentration of 2N.

RESULTS AND DISCUSSION

The results showed that protein isolates obtained from the use of HCl 2N weighing 5,8252 g with a %yield of 11.65 %. Meanwhile, protein isolates obtained using CH3COOH 2N are 5,5931 g with %yield of 11.11 %. In terms of protein content, the Kjeldahl method shows a protein content of 62.48% for HCl 2N and 65.57% for CH3COOH 2N. Whereas based on the Bradford method, the protein content obtained is 3.18% for HCl 2N and 3.33% for CH3COOH 2N.

From this result, it can be concluded that the protein isolates produced with HCl 2N are more efficient than the 2N CH3COOH in terms of the number of isolates obtained. However, the protein content in isolates produced from CH3COOH 2N is higher, both based on the Kjeldahl and Bradford methods. This shows that although HCl 2n is more effective in producing protein isolates, CH3COOH 2N provides higher protein quality.
