Diabetes mellitus is a chronic disease that affects millions of people worldwide, causing various complications, both microvascular and macrovascular. Methodology

This study involved 25 male mice divided into five groups:

Group I: Negative control group, given water only.
Group II: Positive control group, received metformin at a dose of 45 mg/kg body weight (BB).
Group III and IV: Given Kluwih leaf water extract at a dose of 50 mg/kg BW and 100 mg/kg BW, respectively.
Group V: Normal control group, without treatment.

The induction process was carried out through intraperitoneal streptozotocin injection with a dose of 55 mg/kg BW, followed by treatment for 14 days. Measurement of hematological parameters was carried out on the 15th day in the Regional Health Laboratory of North Sumatra Province. To analyze data, the One Way Anova statistical test was used, followed by the Tukey test to determine the difference between the treatment.

The analysis shows that the EADK dose of 50 mg/kg BW is the most effective dose in increasing hematological parameters such as:

Red Blood Cell Count: The number of red blood cells was significantly increased (p = 0.000) in the EADK group compared to the negative control group.
Hemoglobin: Hemoglobin levels were also significantly increased (p = 0.009) in the EADK group.
Hematocrit: Hematocrit levels were significantly increased (p = 0.003) in the EADK group.
Mean Corpuscular Volume: Mean corpuscular volume was significantly increased (p = 0.000) in the EADK group.
Mean Corpuscular Hemoglobin: Mean corpuscular hemoglobin was significantly increased (p = 0.000) in the EADK group.
Platelet Count: Platelet count was significantly increased (p = 0.000) in the EADK group.
Total White Blood Cell Count: Total white blood cell count was significantly increased (p = 0.000) in the EADK group.
Lymphocyte Count: Lymphocyte count was significantly increased (p = 0.021) in the EADK group.

In addition, EADK was also proven to reduce neutrophil levels (p = 0.000) and monocytes (p = 0.001) significantly compared to the negative control group.

Conclusion

Kluwih leaf water extract shows a positive effect on the hematological parameters of diabetes mice. In other words, EADK can help improve hematological conditions, which in turn have the potential to prevent the emergence of various complications associated with diabetes mellitus. This study opens opportunities for further research on the use of kluwih leaves as an alternative therapy in diabetics.
