Introduction

Begomovirus, a type of virus belonging to the Geminivirus group, has become a significant threat to chili plant production in the Karo soil region of North Sumatra. This virus can cause substantial losses to the chili plant, resulting in a decrease in both the quality and quantity of crops. The presence of begomovirus can be difficult to determine visually through morphological symptoms, as the symptoms often resemble those caused by other viruses.

The Importance of Accurate Detection

Accurate detection of begomovirus is crucial for effective disease management in chili plants. The Polymerase Chain Reaction (PCR) technique is a molecular method that can detect the presence of begomovirus with high accuracy. In this study, we aimed to detect the presence of begomovirus in chili plants using the PCR technique.

Materials and Methods

In this study, 20 leaf samples were collected from chili plants that showed symptoms of viral infections from 20 villages in the Karo land area. The samples were then analyzed using the PCR technique with a specific primer for begomovirus. The results showed the presence of morphological symptoms such as vein clearing, thickening and curvature of the leaf bone, and shrinkage on young leaves and leaf strands that are bright yellow.

Results

The PCR process using a specific primer for begomovirus resulted in DNA fragments measuring 1500 bp in all samples of DNA chili plants studied. This indicates that all samples examined were infected by Begomovirus. In addition, anatomical analysis of stomata density showed that healthy chili plants have a higher stomata density compared to plants infected with viruses.

Discussion

The results of this study demonstrate the effectiveness and accuracy of the PCR technique in detecting begomovirus in chili plants. The PCR technique provides a more accurate detection method compared to visual inspection, which often results in false positives or false negatives. With accurate detection, farmers and researchers can take better steps in controlling and managing disease in chili plants, leading to increased production in the Karo land area.
